Human CDK4 Gene Overview

Gene Name cyclin dependent kinase 4
Gene Symbol CDK4
Synonyms CMM3,MCPH31,PSK-J3
NCBI ID 1019
Ensembl ID ENSG00000135446
Uniprot ID P11802
Gene Type Protein coding

Gene Summary

The protein encoded by this gene is a member of the Ser/Thr protein kinase family. This protein is highly similar to the gene products of S. cerevisiae cdc28 and S. pombe cdc2. It is a catalytic subunit of the protein kinase complex that is important for cell cycle G1 phase progression. The activity of this kinase is restricted to the G1-S phase, which is controlled by the regulatory subunits D-type cyclins and CDK inhibitor p16(INK4a). This kinase was shown to be responsible for the phosphorylation of retinoblastoma gene product (Rb). Mutations in this gene as well as in its related proteins including D-type cyclins, p16(INK4a) and Rb were all found to be associated with tumorigenesis of a variety of cancers. Multiple polyadenylation sites of this gene have been reported. [provided by RefSeq, Jul 2008]

Mouse Cdk4 Gene Overview

Gene Name cyclin dependent kinase 4
Gene Symbol Cdk4
Synonyms Crk3
NCBI ID 12567
Ensembl ID ENSMUSG00000006728
Uniprot ID P30285
Gene Type Protein coding

Gene Summary

Enables cyclin binding activity and cyclin-dependent protein serine/threonine kinase activity. Acts upstream of or within several processes, including cellular response to ionomycin; cellular response to lipopolysaccharide; and cellular response to phorbol 13-acetate 12-myristate. Located in bicellular tight junction and nucleus. Part of cyclin D1-CDK4 complex; cyclin D2-CDK4 complex; and transcription regulator complex. Is expressed in several structures, including alimentary system; branchial arch; nervous system; sensory organ; and urinary system. Used to study skin melanoma and type 1 diabetes mellitus. Human ortholog(s) of this gene implicated in several diseases, including carcinoma (multiple); familial melanoma; glioblastoma; obesity; and type 2 diabetes mellitus. Orthologous to human CDK4 (cyclin dependent kinase 4). [provided by Alliance of Genome Resources, Jul 2025]
